Bulking up the hippocampus in schizophrenia: a role for 5-HT1A agonists?

چکیده

SUMMARY The volume of the hippocampus is reduced in patients with schizophrenia, and this deficit tends to become more pronounced with chronicity. Newer antipsychotics may protect against the progressive reductions in hippocampal volume while preliminary data offer hope that specific antipsychotics may act to reverse it. Anxiolytic 5-hydroxytryptamine1A agonists suppress firing activity of dorsal hippocampus CA1 pyramidal neurons through a postsynaptic mechanism: single-unit study in unanesthetized, unrestrained rats.

Recent behavioral studies indicate that conditioned fear response to contextual stimuli is reduced effectively by anxiolytic 5-hydroxytryptame (5-HT)1A agonists. Since the hippocampus seems to play an essential role in associative fear memories evoked by context, it is important to assess the effect of 5-HT1A agonists on pyramidal cell activity in the hippocampus. We examined the effects of 5-H...

Neural Basis for the Ability of Atypical Antipsychotic Drugs to Improve Cognition in Schizophrenia

Cognitive impairments are considered to largely affect functional outcome in patients with schizophrenia, other psychotic illnesses, or mood disorders. Specifically, there is much attention to the role of psychotropic compounds acting on serotonin (5-HT) receptors in ameliorating cognitive deficits of schizophrenia. It is noteworthy that atypical antipsychotic drugs (AAPDs), e.g., clozapine, me...

Perspectives in Pharmacology Improving the Treatment of Schizophrenia: Focus on Serotonin (5-HT)1A Receptors

Although antagonism of mesolimbic dopamine D2 receptors by neuroleptics such as haloperidol attenuates positive symptoms of schizophrenia, a significant population of “resistant” patients fails to respond while negative and cognitive symptoms are little modified. Furthermore, concomitant blockade of striatal D2 receptors is associated with extrapyramidal motor side effects. The superior “atypic...

P2: The Role of Neuronal Nitric Oxide Synthase on the Anti-Seizure Effects of 5-HT1A Receptors in Perforant Pathway Kindling Model in Rat

Neuronal nitric oxide synthase (nNOS) plays a role in synaptic potentiation and kindling process. The relationship between nNOS and 5-HT1A receptors also nearly has been specified. In this research,we investigate the role of nNOS on the anticonvulsant effect of 5-HT1A receptors. 24 male (280 ± 30 g) were randomly assigned to four groups (vehicle,NI,Way 100635 and NI + Way100635) (n = 6). ...

Does stimulation of 5-HT(1A) receptors improve cognition in schizophrenia?

Cognitive impairment is a key feature of schizophrenia and may be the most important determinant of outcome in schizophrenia. This impairment is diffuse and may reflect abnormalities in frontal cortex, hippocampus and other brain regions. While deficits in glutamatergic, GABAergic, dopaminergic and cholinergic impairment have received the most attention as the basis of this impairment, there ar...
